Proposta de Modificação da Biossíntese da Lignina como Estratégia para Correcção de Defeitos em Madeiras

Sumário.A lignina corresponde a cerca de 15 a 35% do peso seco da madeira e assume papel importante na estrutura da parede celular, e pode ser modificada facilmente através da engenharia genética, ou por outra técnica que confira modificações sobre as actividades das enzimas PAL, 4CL, C4H, C3H, OMT, F5H, CCR, CAD, que regulam as reacções da etapa que antecede a polimerização da lignina. Através destas, as plantas transformam lignina siringílica que é mais frágil estruturalmente (IFM maior), em lignina guaiacílica, a qual se mostra estruturalmente muito mais ramificada e rígida (IFM menor). A interferência no controle biossintético da lignina a nível da organização estrutural via técnicas biomoleculares pode resolver possivelmente alguns problemas congénitos de algumas espécies madeireiras de interesse comercial, como é o caso do Eucalipto que apresenta rachaduras após a colheita. Considerando que a resistência da parede celular depende da natureza estrutural da lignina, a biossíntese de ligninas modificadas em termos da definição da arquitectura molecular dentro da parede celular abre novas perspectivas em busca de soluções para maior aplicabilidade da madeira na indústria. Desta forma, o objectivo deste artigo foi propor soluções para alguns defeitos naturais da madeira modificando a rota biossintética da lignina Guaiacílica/Siringílica à lignina Cumarílica/Guaiacílica, com base em técnicas de bioquímica e engenharia genética.

Abstract. Lignin, which corresponds to approximately 15 to 35% of the dry weight of the wood, works as a binding agent for cell wall, and could easily be modified by genetic engineering or by other experimental techniques, affecting the activity of the PAL, 4CL, C4H, C3H, OMT, F5H, CCR, CAD enzymes, which mediate reactions before lignin polymerization. Such techniques will allow plants to change syringyl lignin, which is more fragile (IMF higher), into guaiacyl lignin, which is much more rigid (IMF lower). This opens new perspectives for the control of cracks as observed in Eucalyptus wood after harvest. Considering that the strength of the cell wall depends on the structural lignin organization, the biosynthesis of modified lignin in terms of molecular architecture within the cell wall opens up new possibilities in the search for solutions for greater applications of wood in industry. In this way, the scope of this article is to propose solution to solve some natural wood defects, suggesting changes in the biochemical lignin pathway which transforms Guaiacyl/Syringyl lignin into Coumaryl/Guaiacyl lignin.

Résumé.La lignine représente environ 15 à 35% du poids sec du bois et est très importante dans la structure de la paroi cellulaire. Elle pourrait être facilement modifiée grâce à la génétique ou par d'autres techniques expérimentales, affectant l'activité de PAL, 4CL, C4H, C3H, OMT, F5H, CCR, DAO, qui règlent les réactions avant polymérisation de la lignine. Grâce à celles-ci la plante transforme la lignine de syringil, qui est plus fragile (IFM plus élevé), lignine de guayacil, qui est beaucoup plus rigide (IFM plus bas). Ce travail peut résoudre le problème de quelques espèces à bois d'intérêt commercial, comme c'est le cas pour l'eucalyptus qui présente des fentes après récolte. Considérant que la résistance de la paroi cellulaire dépend de la nature structurale des lignines, la biosynthèse de lignines modifiées ouvre de nouvelles perspectives pour obtenir des solutions de meilleure applicabilité dans l'industrie du bois, basée sur la définition structurale de l'architecture de la paroi cellulaire. De cette façon, l'objectif de cet article est de proposer des solutions pour quelques défauts naturels du bois en transformant le chemin biosynthétique de la lignine, avec comme base des techniques de biochimie et du génie génétique, transformant le Guayacil/Syringil en lignine de Coumaril/Guayacil.
